Plant innate immunity is a potential surveillance system of plants and is the first line of defense against invading pathogens. The immune system is a sleeping system in unstressed healthy plants and is activated on perception of the pathogen-associated molecular patterns (PAMP; the pathogen’s signature) of invading pathogens. The PAMP alarm/danger signals are perceived by plant pattern-recognition receptors (PRRs). The plant immune system uses several second messengers to encode information generated by the PAMPs and deliver the information downstream of PRRs to proteins which decode/interpret signals and initiate defense gene expression. Activation of the ‘sleeping’ plant innate immune system by using different biotechnological tools would suppress the development of a wide range of plant pathogens in economically important crop plants. Enhancement of disease resistance through altered regulation of plant immunity signaling systems would be a durable and publicly acceptable technology in plant disease management. This book describes the most fascinating PAMP-PRR signaling complex and signal transduction systems. It also discusses the highly complex networks of signaling pathways involved in transmission of the signals to induce distinctly different defense-related genes to mount offence against different biotrophic, hemibiotrophic, and necrotrophic pathogens.

Filaggrin proteins are expressed in several human tissues, including the skin, oral mucosa, conjunctivae, esophagus, cervix, and testes. While filaggrin guarantees both structural and functional normality of the human epidermis, its role in other tissues is largely unknown. Epidermal deficiency of filaggrin causes dry and scaly skin, but also strongly increases the risk of skin diseases, in particular ichthyosis vulgaris and atopic dermatitis. Importantly, the risk of related asthma, hay fever, and food allergy is also increased although filaggrin is not expressed in the airways and gastrointestinal tract. Common FLG mutations, leading to reduced filaggrin expression, have mainly been identified in Europeans and Asians, reaching a prevalence of 5-10%. Even in the absence of atopic dermatitis, the skin of individuals with FLG mutations differs fundamentally from normal skin, for example by generating more vitamin D and allowing chemicals and allergens to more widely penetrate across the stratum corneum. This textbook provides comprehensive and detailed coverage of the effects of filaggrin and filaggrin gene (FLG) mutations in health and disease (cutaneous and non-cutaneous) and also discusses the basic science, epidemiology, management, and future research areas. Il existe une véritable « blessure de l’identité » dans l’épreuve de la maladie : une dégradation du sentiment de soi et de l’estime personnelle qui atteignent très profondément le sujet malade une catastrophe intime, en somme. Les modifications corporelles et psychiques que la maladie provoque, la difficulté à se reconnaître dans des postures ou des traits inconnus, entretiennent un sentiment d’étrangeté ou d’exil hors de soi. Il faut donc s’interroger sur une forme de soin qui s’attache à réconcilier le patient avec une identité transformée par la maladie, le familiariser avec une nouvelle existence, l’aider à retrouver le goût de soi. Cette « peau de soins » embrassant un patient écorché par la violence d’une médecine souvent intrusive doit ainsi permettre de réparer une identité blessée.
